Evaluation of Mrna Expression Profiling of NF-?B Mediated Signaling in COPD Patients

Introduction: Chronic Obstructive Pulmonary Disease (COPD) will become 3rd prominent cause of mortality and morbidity globally by 2030. NF-κB mediated inflammatory responses perform a significant role in the progress of COPD by regulating the expression of pro-inflammatory cytokines and chemokines of immune cells. Aim and Objectives: To assess the expression of NF-κB gene and inflammatory markers (interleukin (IL) - 1β and IL-8) in COPD cases and healthy controls. Methods: In this case-control study, peripheral blood was isolated from 60 COPD cases and healthy controls, each. The serum level of interleukin-1β as well as IL-8 were measured by ELISA in both groups. mRNA expression of NF-κB p65, IL-1β and IL-8 were assessed in blood samples by qRT-PCR. Results: We found that serum levels of IL-1β (7.6 ± 2.42 pg/mL, 2.9±0.64 pg/mL, p < 0.001) and IL-8 (74.32±12.69 pg/mL, 41.27±10.4 pg/mL, p < 0.001) were significantly raised in COPD cases in comparison to healthy controls, respectively. The mRNA expressions of NF-κB p65 (p =0.005), IL-1β (p < 0.001) along with IL-8 (p < 0.001) were upregulated in COPD patients. Conclusion: Significant upregulation of NF-κB p65 mediated inflammation and its association with severity in COPD patients showed that NF-κB p65 may act as a potential biomarker for lung health deterioration.
